Study of the distribution of 18 elements in Rauvolfia Verticillate by inductively coupled plasma optical emission spectrometry

Abstract: A rapid and sensitive inductively coupled plasma optical emission spectrometry (ICP-OES) method was developed to determine simultaneously for the first time 18 elements (Mg, Ni, Na, K, Ca, Ba, Mo, Mn, Cr, Fe, Cu, Li, Zn, Co, V, Cd, As and Pb) which associated with hypertension in the traditional Chinese drug—Rauvolfia verticillate. The distributions and concentrations of the 18 elements in Rauvolfia verticillate were measured after digestion of a powdered sample (approximately 1.0 g) with violet acid (HNO3/HClO4 : 4/1, v/v). The method was shown to be sensitive and its limit of detection (0.335-25.44 μg·L-1), and limit of quantification (1.12-84.80 μg·L-1) were established. The recovery of all the elements tested was more than 82.74 %, and the method exhibited an RSD of 0.2 %-14.1 %. The results obtained explained a possible relationship between the distribution of elements in the whole plant and its pharmacodynamic action as well as environmental factors involved in the growth process. Moreover, this work also contributed to the quality control standard of traditional Chinese drugs with regard to the elements involved.
