Quantitative Determination of Aristolochic Acid in Asarum heterotropoides Fr. Schmidt var. mandshuricum (Maxim.) Kitag.

Abstract: A reversed-phase high performance liquid chromatographic method was established for the quantitative determination of aristolochic acid
in different parts of Asarum heterotropoides Fr. Schmidt var. mandshuricum (Maxim.) Kitag. (Chinese herbal name: Liaoxixin) produced in
different places and growing years. Aristolochic acid was successfully separated on a Diamonsil C18 column(5μm, 200×4.6 mm). The mobile
phase was a mixture of methanol, water and glacial acetic acid (73:26:1 v/v/v), with isocratic elution at a flow rate of 1.0 mL/min. Detection
was carried out at 390nm. The compound was identified by comparing its retention time and UV spectra in the 200-400nm region with an
authentic standard. Regression equations showed there was a good linear relationship between the peak areas of the constituent and its
concentrations (correlation coefficients: 0.9999).The prosposed method has been successfully applied to the quantitative determination of
aristolochic acid in Liaoxixin and requires only 20 min. The content of aristolochic acid in different parts of Liaoxixin produced in different
places and growing years has been examined by HPLC. There is no or only a small amount of aristolochic acid in the underground parts,
while most is in the overground parts. As the growing year increases, the content of aristolochic acid decreases although the place of
production changes. There is a variation in the content of aristolochic acid in Liaoxixin produced in different places and different growing
years.
