Simultaneous determination of five flavonoids in Herba Euphorbiae Humifusae by high-performance liquid chromatography

Abstract: A high-performance liquid chromatographic (HPLC) method was developed for the simultaneous determination of five flavonoid glycosides, quercetin-3-O-galactosyl-6″-O-gallate (E1), luteolin-7-O-β-D-glucoside (E2), quercetin-3-O-β-D-glucoside (E3), apigenin-7-O-β-D-glucoside (E4) and kaempferol-3-O-β-D-glucoside (E5), contained in Herba Euphorbia Humifusa. The samples were separated on a Phenomenex C18 column (250mm×4.6 mm, 4μm) with a mobile phase of methanol and water containing 0.5 % glacial acetic acid (42.5:57.5, v/v) at a flow rate of 0.9 ml· min-1 and detected at 330 nm. The five compounds were completely separated within 35 min. All calibration curves showed good linearity (r>0.999) within the test ranges, E1: 7.60-304 μg·ml-1 (r=1), E2: 1.04-41.6 μg·ml-1 (r=0.9999), E3: 12.6-504 μg·ml-1 (r=0.9997), E4: 0.82-32.8 μg·ml-1 (r=0.9997) and E5: 2.40-96.0 μg·ml-1 (0.9998), respectively. Also, the precision, repeatability and stability were all less than 3.0 % RSD and the recoveries were between 97.4 % and 100.8 %. The method is simple, accurate, and very reproducible. It was successfully applied to the analysis of 19 samples, six of which were collected in Shenyang at different developmental stages. The results indicated that the HPLC assay is very suitable as a quality control method for Herba Euphorbiae Humifusae.
