    Biological characteristics of a pathogenic bacterium in Panax notoginseng

    Root rot of Panax notoginseng is the most serious disease affecting its cultivation, which may be affected by its rhizosphere microbial community. We collected rhizosphere soil samples from Panax notoginseng suffering from root rot disease and isolated a strain designated SYP-B552. In the present study, strain SYP-B552 was found to be correlated with root rot disease and increased mortality of Panax notoginseng. Cells were Gram-negative, aerobic, non-motile and short rod shaped. The optimal growth of the strain occurred at 28°C and pH 7.0. The highest 16S rRNA gene sequences similarities between the strain SYP-B552 and other Chryseobacterium type strains were 98.10%. The predominant isoprenoid quinone was MK-6, and the major fatty acids were iso-C15:0 (40.07%) and Summed Feature 9 (comprising C16:0 10-methyl, 19.53%). The G+C content of the genomic DNA was 31.3 mol%. On the basis of the phylogenetic and biochemical data, strain SYP-B552 represents a species of the genus Chryseobacterium. Strain SYP-B552 could be regarded as pathogenic bacteria for exploring effective control measures for root rot disease in Panax notoginseng.

    Determination of the Total Content of Iodine in Wuhai Yingliu Pill and Neixiao Luoli Pill and Related Drugs within Them by UV-Vis spectroscopy

    A method for determination of total content of iodine in Wuhai Yingliu Pill and Neixiao Luoli Pill was developed by UV-Vis spectroscopy at 420 nm wavelength. Firstly, all the sample solutions in this work and the mixture of K2CO3-ZnSO4 and KClO3- NaCl were heated in dry bath at 300°C for about 3 h; Then the supernatant was collected after dissolving with water; At last, the absorbance was measured based on the catalytic of iodine in arsenic cerium oxide reduction reaction. Using this method, 23 samples were analyzed. A good linear regression relationship was obtained over the range of 0-1200 μg/L (r=0.9989) for standard solutions, the recovery rate was 96.54%, RSD=3.02% (n=9). The method was successfully applied to analyze these two Chinese patent medicines as a quantity control for clinical use in different types of thyroid disease.

    Mutagenesis of a newly isolated nitrilaseproducing strain and gene cloning and expression of nitrilase from Agrobacterium sp

    In our previous work, new wild-type strain (WT16) that exhibited nitrilase activity toward 3-cyanopyridine was isolated from soil through enrichment culture using acetonitrile or 3-cyanopyridine as the sole source of nitrogen. To improve the strain’s nitrilase activity for industrial application, it was further subjected to mutagenesis using LiCl, and the mutant strain Agrobacterium sp. SPUA14 was obtained. The nitrilase activity of Agrobacterium sp. SPUA14 was markedly increased by 50.6% compared with WT16 (26.23 μmol·g-1·h-1). The nitrilase from Agrobacterium sp.SPUA14 was subsequently amplified and expressed in E. coli BL21 (DE3). The apparent Km and rmax of the nitrilase against 3-cyanopyridine were 8.35×10-2 mol·L-1 and 3.575× 10-3 mol·L-1·min-1, respectively. After 40 min reaction using E. coli BL21/pET22b-a14 resting cells, 3-cyanopyridine at the concentration of 10 mmol·L-1 was completely hydrolyzed and 91.2% conversion rates were attained. The determination of the substrate tolerance indicated that the optimum substrate concentration of nitrilase expressed in the engineered bacteria is approximately 300 mmol·L-1, which makes it a potential biocatalyst for the production of nicotinic acid.
