Analysis of the compounds present in rat blood after oral administration of the extract of Gushudan (a Chinese compound prescription)

Abstract: Gushudan is a “kidney-tonifying” and “bone-strengthening” Chinese compound prescription. In order to investigate its active constituents, extracts of Gushudan and its principle constituent, Herba Epimedii, were administered orally to rats. Then the rats’ serum was analyzed by HPLC-UV and UPLC-MS. The HPLC and UPLC/MS profiles of Gushudan extract, blank serum, and rat serum after oral administration of Gushudan extract and Herba Epimedii extract were compared. Some constituents existing in the prescription such as epimedin B, icariin and osthole and unidentified metabolites were detected in the rat sera after administration of the Gushudan extract. Therefore, epimedin B, icariin, osthole and other metabolites were systemically absorbed and could be responsible for the biological activity of the Gushudan prescription.
