Simultaneous Determination of genistein, genistin, daidzein, daidzin, glycitein and glycitin in different fermented products of Semen sojae praeparatum.

Abstract: A simple and reliable ultra performance liquid chromatography method was established for determination of genistein, genistin, daidzein, daidzin, glycitein and glycitin content in different period fermentation of Semen sojae praeparatum. The chromatographic separation was performed on an ACQUITY UPLC BEH C18 (2.1*50 mm, 1.7 μm). The mobile phase was composed of acetonitrile (A), 0.1% formic acid in water (B). The gradient elution conditions were as follows: 0-8 min, 10-24% (A), 8-10 min, 24-50% (A), 10-12 min, 50% (A). The chromatogram was monitored at wavelength of 260 nm during the experiment. The result showed that the content of daidzin, glycitin and genistin were reduced, however the content of daidzein, glycitein and genistein were increased. The method is simple, accurate and reproducible. It can be used to provide the theory basis for the quality control of traditional Chinese medicine Semen sojae praeparatum. Finally, it provides an identification standard for production and market circulation of Semen sojae praeparatum.
