Determination of (-)-secoisolariciresinol 4-O-β-Dglucopyranoside in Glehnia littoralis by SPE-HPLC

Determination of (-)-secoisolariciresinol 4-O-β-Dglucopyranoside in Glehnia littoralis by SPE-HPLC

摘要: A new simple and reliable HPLC method was established for determining the contents of (-)-secoisolariciresinol 4-O-β-Dglueopyranoside in Glehnia littoralis. HPLC analysis was performed on Classic C18-SPE column (500 mg, 6 ml) and C18 column (200 mm×4.6 mm, 5 μm) using a gradient elution of water (A) and methanol-acetonitrile (B, 10:90, v/v), the column temperature was set at 30°C, the flow rate was 1.0 ml·min-1 and the detection wavelength was at 230 nm. The linear range was 10.7- 214.0 μg·ml-1, the average recovery was 96.1%, and RSD was 1.2%. The method was reliable, accurate, and was successfully applied to quantify the (-)-secoisolariciresinol 4-O-β-D-glucopyranoside content in Glehnia littoralis collected from different areas in China.
