    A systematic quantitative method for quality evaluation of Bupleurum chinense DC. based on multi-wavelength fusion fingerprint combined with multivariate chemometrics

    A quantitative evaluation method for the quality consistency of Bupleurum chinense DC. was developed by combining multiwavelength fusion fingerprint with chemometric methods. The fingerprint information of Bupleurum at five wavelengths (210, 254, 265, 280 and 320 nm) collected by HPLC-DAD instrument was fused to overall control its quality. The fusion fingerprints and four components (saikosaponin a, saikosaponin d, rutin, quercitrin) were studied by systematic quantitative fingerprint method and principal component analysis, respectively. Besides, fingerprint-efficacy relationship was constructed in vitro using partial least-squares model. In conclusion, all work can serve as a valid reference for the quality control of Bupleurum.

    A validated reversed phase HPLC method for determination of process-related impurities in DHDK-loaded mPEG-PLA micelles

    A new compound named DHDK was isolated from Viscum coloratum and identified as (1E,4E)-1,7-bis(4-hydroxyphenyl)hepta -1,4-dien-3-one. DHDK, a new potential anti-tumor drug, was synthesized and prepared poly(ethylene glycol)-poly(D.L-lactic acid) (mPEG-PLA) micelles due to cytotoxic effects on a range of cancer cells. A revered phase liquid chromatographyic method has been developed and validated for the determination of potential related impurities in DHDK-loaded mPEG-PLA micelles. The micelles was subjected to the stress conditions of acid, base, oxidation, thermal and photolysis. The determination was achieved on Phenomenex SynergiTM 4 μm Fusion-RP 80Å (250 mm × 4.6 mm, 4 μm) with a gradient elution and the detection wavelength was set at 220 nm. Regression analysis shows as r value (correlation coefficient) of greater than 0.999 for DHDK and six potential impurities. Recoveries of impurities were found between 90.0% and 108.0%. The developed RP-LC method was validated with respect to linearity, accuracy, precision and robustness, which can be used for determination of related impurities in DHDK-loaded mPEG-PLA micelles.

    Insulin and Leptin resistance are the hallmarks for the development of type 2 diabetes mellitus (T2DM) and obesity. Both of these conditions are associated with increased activity and idiom of Protein Tyrosine Phosphatase 1B (PTP 1B). Therefore, inhibition of PTP 1B activity or down-regulation of its expression has become a striking area of therapeutics development in order to ameliorate the phenomenon of Insulin and Leptin resistance. Lack of awareness and late diagnosis makes diabetes difficult to manage. PTP 1B inhibitory activity by Erythroxylum monogynum plant extracts in the cytosolic supernatant of goat skeletal muscle and the liver was estimated. Erythroxylum monogynum plant extraction was done in hydroalcoholic solution (water–ethanol 20:80 (v/v)) and Methanol. The Polyphenols and protein content were identified in the hydroalcoholic and methanolic extracts. The greatest PTP 1B inhibitory activity in goat liver and skeletal muscle was found in the hydroalcoholic extract. Erythroxylum monogynum is a potent natural source of PTP 1B inhibition and may become cost-effective therapeutics in correcting metabolic disturbances of T2DM, obesity and cardiovascular disorders.

    Studies on the Chemical Components and Biological Activities of the genus of Caragana

    The genus Caragana is a member of the family of Fabaceae, subfamily Faboideae and there are 62 species, 9 varieties, 12 deformations grown in China. More than 200 components have been identified as flavonoids, stilbenoids, which are the major constituents of this genus, and phenylpropanoids, terpenoids, alkaloids also have been isolated from this genus. Modern pharmacological studies suggest diverse pharmacological activities of the herbals from the genus, including anti-tumor, antiinflammatory, anti-virus, and anti-oxidant activities. This review summarized the update research progress on the chemical components and pharmacological effects of the Cragana genus.
